How much of a small business can realistically be automated?

The short answer

There is no honest percentage, and anyone quoting one is guessing. What can be measured is which processes qualify: repeatable, rule-bound, high-touch and error-prone ones are candidates, volume ranks them, and anything needing taste or legal judgement keeps a person at one gate.

How can we tell whether a process is ready to automate?

Check that its rules are agreed. Run any disputed rule by hand for a fortnight: if people follow it and nothing breaks, automate it; if they quietly go around it, you have learned something worth more than the automation would have been.

Should we automate the process we do most often?

Not automatically. Frequency is a multiplier, not a reason: a process run constantly has met more exceptions than anyone has written down. Qualify it on the four signals first, then let volume decide the order.
